The primary objective of the study was to evaluate and investigate the reliability of multi-colour fluorescence in-situ hybridization (FISH) technique and comparative genomic hybridization (CGH) in the cytogenetic analysis of non-Hodgkin's lymphoma cell karyotypes. Cytogenetic investigations were attempted on serious of lymph node biopsies from NHL patients and five human lymphoma cell lines. Multi-colour FISH development using advance CCD camera and computer software allowed the identification of a novel translocation in two cases of NHL patients and for a more detailed understanding of karyotype abnormalities in both established cell lines and direct preparations of NHL samples. Using CGH, I identified novel genetic abnormalities affected by gain or loss of genetic material in most of the NHL cases and some of the cell lines. In the present study, CGH, cytogenetics and multi-colour FISH were found to be complement each other. This could focus attention on specific chromosomes for future molecular biological studies. The future combination of these approaches may contribute to an increased understanding of malignant lymphoma progression and refine patient diagnosis.
